The verse you referred to reads thusly in its context:

John 6:
37 All that the Father giveth me shall come to me; and him that cometh to me I will in no wise cast out.
38 For I came down from heaven, not to do mine own will, but the will of him that sent me.
39 And this is the Father's will which hath sent me, that of all which he hath given me I should lose nothing, but should raise it up again at the last day.
40 And this is the will of him that sent me, that every one which seeth the Son, and believeth on him, may have everlasting life: and I will raise him up at the last day.

To get a good grasp on my answer, you need to study booklet two, chapter five, of our on-line book THE CLOCK OF THE AGES. In this chapter you will find a discussion on the "peaks of prophecy" wherein we point out the events prophesied for each "peak" and how they will effect those who will be involved. As per the illustration below, taken from the chapter five of the book, you can see that each peak of prophecy ends an age: i.e., the first peak ended the Age of the Law, the second peak will end the Age of Grace in which we now live, and the third peak will end the Kingdom Age yet to come.

If you will look at the illustration, you will notice that the last valley is labelled "THE MILLENNIUM." However, the Millennium is know by other names. It is also known in the church as the 1000 years of peace because of the following Scriptures.

Revelation 20
1 And I saw an angel come down from heaven, having the key of the bottomless pit and a great chain in his hand.
2 And he laid hold on the dragon, that old serpent, which is the Devil, and Satan, and bound him a thousand years,
3 And cast him into the bottomless pit, and shut him up, and set a seal upon him, that he should deceive the nations no more, till the thousand years should be fulfilled: and after that he must be loosed a little season.
4 And I saw thrones, and they sat upon them, and judgment was given unto them: and I saw the souls of them that were beheaded for the witness of Jesus, and for the word of God, and which had not worshipped the beast, neither his image, neither had received his mark upon their foreheads, or in their hands; and they lived and reigned with Christ a thousand years.
5 But the rest of the dead lived not again until the thousand years were finished. This is the first resurrection.
6 Blessed and holy is he that hath part in the first resurrection: on such the second death hath no power, but they shall be priests of God and of Christ, and shall reign with him a thousand years.

The Millennium is also known as the Kingdom Age because Jesus will establish His Kingdom for the 1,000 years and rule the earth with a rod of iron (Rev. 2:27).

Also, the Millennium will be known as the Sabbath. Let's look at some Scripture.

II Peter 3:8
But, beloved, be not ignorant of this one thing, that one day is with the Lord as a thousand years, and a thousand years as one day.

In our book THE CLOCK OF THE AGES, we discuss in some detail how that in the prophetic time of the Bible, a thousand years is as one day. We also study the geneaology of the Bible and the time line of the Bible, demonstrating how that it has been some 6,000 years now since the fall of Adam and Eve in the Garden of Eden. (This is not to say that the Bible teaches that the earth is only 6 thousand years old, for it could have existed for eons before the first couple fell into sin, taking the human race with them.) Consequently, the final prophetic day will be the 1,000 years of the Kingdom Age which will also be know as the Sabbath wherein the Lord will reign with a rod of iron.

Matthew 12:8
For the Son of man is Lord even of the sabbath day.

With this we can establish the day that Jesus was talking about in the Scripture you referred to: i.e.,

John 6:
...39 And this is the Father's will which hath sent me, that of all which he hath given me I should lose nothing, but should raise it up again at the last day.
40 And this is the will of him that sent me, that every one which seeth the Son, and believeth on him, may have everlasting life: and I will raise him up at the last day.

So, the day that Jesus was talking about will be the final prophetic day or the Lord's Sabbath day wherein He will rule the earth from Jerusalem. But we should also note as per the above Scripture, that Jesus included the short period of Great Tribulation/Wrath just prior to His Kingdom Age in the "prophetic day.": Likewise, the similar short period of Great Tribulation/Wrath at the end of the Millennium is included in the final "prophetic day."

Now, to finish answering your question, we need to note that the people who lived (will live) in the valleys (ages) before each peak (see the illustration above) were given certain conditions that had to be met to be accepted of God. Those who lived during the Age of the Law had to fear/trust God, to do their best to keep the law given to Moses, to observe the rituals given in the law and to, by faith, offer blood sacrifices as per the law. For those who live during our age, God has commanded all men (and women) to repent of their sins and to believe in the death, burial, and resurrection of Jesus Christ, His only Begotten Son. For the people who will live during the Kingdom Age, we're given little in the Word as to the conditions that must be met to be accepted by their Maker--but remember, Jesus is the Word and He will be living with them at the time; so, they'll know what is required of them. We do know from the Scriptures that whatever the conditions, as with the other ages, there will be some living during the Kingdom Age who will accept Jesus while there will be others who will be deceived by Lucifer (Satan) to the eternal destruction of their souls. Many of those who will believe in Jesus during the final prophetic day will become martyrs for the Lord. We just read in Revelation 20:4 where those who will be martyed in the short period after the Rapture of the Church and and just prior to the Lord's return with His saints to establish His Kingdom will be resurrected to rule with Him during the 1,000 years. Likewise, those who will be martyred at the end of the Kingdom Age will be resurrected at that time to receive a martyr's reward. Take note, though, that the time of rewards for the Kingdom Age saints will not be the same as the time of judgment for the lost. The lost of all the ages will have their own time to stand before their Maker to give account for all that they did while in this flesh--the day of their future judgment is known as the Great White Throne Judgment-- and no believer will have to appear at this judgment for Jesus took the judgment of all believers upon Himself as He hung upon the cross of Calvary.

II Corinthians 5:17-21
Therefore if any man [be] in Christ, [he is] a new creature: old things are passed away; behold, all things are become new.
And all things [are] of God, who hath reconciled us to himself by Jesus Christ, and hath given to us the ministry of reconciliation;
To wit, that God was in Christ, reconciling the world unto himself, not imputing their trespasses unto them; and hath committed unto us the word of reconciliation.
Now then we are ambassadors for Christ, as though God did beseech [you] by us: we pray
[you] in Christ's stead, be ye reconciled to God.
For he hath made him [to be] sin for us, who knew no sin; that we might be made the righteousness of God in him.

And this was done as was prophesied in the Old Testament

Isa 53:11
He shall see of the travail of his soul, [and] shall be satisfied: by his knowledge shall my
righteous servant justify many; for he shall bear their iniquities.

If you haven't taken the time to read all that's in the book, THE CLOCK OF THE AGES, you might do so. I think that you might be pleasantly surprised at what's in it.
